چکیده

Glibenclamide has beneficial effects in animal models of cerebral ischemia. The cellular targets of glibenclamide are proposed to be neurons, endothelial cells, oligodendrocytes and astrocytes. However, independent studies have shown that the SUR1-formed ATP-dependent potassium (KATP) channels are expressed by microglia, which reinforces the idea that glibenclamide may also target microglia and modulate their inflammatory phenotype. This comment to ‘Sulfonylurea receptor 1 in central nervous system injury: a focused review’ provides new insights on the putative role of the microglial KATP channel in mediating, at least in part, the neuroprotective and neurorestorative effects of glibenclamide after stroke.

Glibenclamide enhances neurogenesis and improves long-term functional recovery after transient focal cerebral ischemia.

Glibenclamide is neuroprotective against cerebral ischemia in rats. We studied whether glibenclamide enhances long-term brain repair and improves behavioral recovery after stroke. Adult male Wistar rats were subjected to transient middle cerebral artery occlusion (MCAO) for 90 minutes. A low dose of glibenclamide (total 0.6 μg) was administered intravenously 6, 12, and 24 hours after reperfusio...

ATP-dependent potassium channel blockade strengthens microglial neuroprotection after hypoxia-ischemia in rats.

Stroke causes CNS injury associated with strong fast microglial activation as part of the inflammatory response. In rat models of stroke, sulphonylurea receptor blockade with glibenclamide reduced cerebral edema and infarct volume. We postulated that glibenclamide administered during the early stages of stroke might foster neuroprotective microglial activity through ATP-sensitive potassium (K(A...

P171: Microglia Cell, Major Player in the Central Nervous System Inflammation

Inflammation, a self-defensive reaction against various pathogenic stimuli, may become harmful self-damaging process. Increasing evidence has linked chronic inflammation to a number of neurodegenerative disorders including alzheimer's disease (AD), parkinson's disease (PD), and multiple sclerosis (MS). In the central nervous system, microglia, the resident innate immune cells play major role in...

Blockade of microglial KATP -channel abrogates suppression of inflammatory-mediated inhibition of neural precursor cells.

Microglia positively affect neural progenitor cell physiology through the release of inflammatory mediators or trophic factors. We demonstrated previously that reactive microglia foster K(ATP) -channel expression and that blocking this channel using glibenclamide administration enhances striatal neurogenesis after stroke. In this study, we investigated whether the microglial K(ATP) -channel dir...

Stroke induces rapid activation and expansion of microglia, but the main source of microgliosis is controversial. Here we investigated the formation of microgliosis and infiltration of circulating cells in a photothrombosis stroke model by taking advantage of parabiosis and two-photon microscopy. We found that a small population of blood-derived CX3CR1 + cells infiltrated the cerebral parenchym...
